摘要

Abstract

To address issues related to identifying plum varieties and their complicated genetic relationships,the genetic diversity and relationships of 11 Prunus mume varieties from Yongtai,Xianyou,Xiuyu,and Zhao'an in Fujian Province were analyzed using ISSR molecular marker technology.The results showed that 22 polymorphic primers were selected from 100 ISSR primers,amplifying a total of 91 gene loci,of which 72 were polymorphic,representing 79.12%of the total loci.Popgene analysis revealed that the mean genetic diversity index(Nei's)was 0.257 6,the mean number of effective alleles was 1.425 7,and the mean Shannon's information diversity index was 0.393 3.The UPGMA cluster analysis showed that the 11 varieties could be divided into three categories.The first major category is'Dengjiashan No.1''Dengjiashan No.2''Xiaoxicun No.3'and'Baifenmei'.The second category includes'Xiaoxicun No.1''Xiaoxicun No.2''Hongmei''Dashuimei''Qingmei No.1 liziye'and'Wengzhong'.The third category only has one variety,'Yuanshengzhong'.The genetic consistency of the 11 varieties ranged from 0.615 4 to 0.824 2,and the genetic distance ranged from 0.193 4 to 0.485 5.The genetic consistency between'Xiaoxicun No.1'and'Xiaoxicun No.2'in Yongtai,was the highest,with the smallest genetic distance,indicating the closest genetic relationship.In contrast,the genetic uniformity of'Dengjiashan No.1'from Yongtai was the lowest,with the largest genetic distance,indicating the farthest genetic relationship.This study confirmed the reliability of ISSR molecular marker technology in analyzing genetic diversity and relationships of P.mume varieties,and provided scientific and technical insights for addressing the problem of distinguishing P.mume varieties among excellent fruit trees in southern China.Additionally,this study offers a theoretical basis for promoting the cultivation of different P.mume varieties in Fujian Province.
