木麻黄CRY1基因的克隆及生物信息学分析
Cloning and Bioinformatics Analysis of CRY1 Gene in Casuarina equisetifolia

Abstract
Cryptochrome is an important blue light receptor affecting the plant growth.In order to explore the mechanism of cryptochrome protein of Casuarina equisetifolia in the light regulation network,the CRY1 gene sequence of Casuarina equisetifolia was screened and identified from the Casuarina equisetifolia genome database and named CeCRY1.The full-length cDNA sequence of CeCRY1 gene from Casuarina equisetifolia was cloned by PCR experiment,and the CeCRY1 protein was transiently expressed in tobacco cells by agrobacterium-mediated transformation method,thus to carry out the subcellular localization analysis,and then the bioinformatics analysis of CeCRY1 protein was performed.The results showed that the full-length cDNA sequence of CeCRY1 gene from Casuarina equisetifolia was 2 046 bp,and the number of amino acids encoded by CeCRY1 gene was 681.The molecular weight of the protein was 76.60 kDa,and the isoelectric point PI was 5.64.The subcellular localization analysis showed that CeCRY1 protein was localized in the nucleus and cytoplasm of tobacco cells.The phylogenetic tree analysis showed that CeCRY1 protein was closely related to CRY1 proteins in some species of Salicaceae and Morus notabilis of Moraceae.关键词
