红景天苷对低氧诱导大鼠心肌细胞凋亡及氧化应激损伤、炎症反应的影响
Effects of salidroside on apoptosis,oxidative stress injury and inflammation of cardiomyocytes induced by hypoxia in rats

Abstract
Objective To investigate the effects of salidroside(SAL)on hypoxia-induced apoptosis,oxidative stress injury and inflammatory response in rat cardiomyocytes.Methods H9C2 rat cardiomyocytes were used.The experiment was divided into normoxia group(21%O2)and hypoxia group(2%O2).Cardiomyocytes in the hypoxia group were first treated with hypoxia for 24 hours in a dedicated hypoxia incubator,then drugs were added,and then they were treated under hypoxia for another 24 hours.Cell viability was detected by CCK-8 kit,the degree of oxida-tive stress injury was observed by detecting lactate dehydrogenase(LDH)activity,the levels of inflammatory factors interleukin(IL)-1β and IL-6 were determined by enzyme-linked immunosorbent assay(ELISA),and the relative ex-pression levels of apoptosis-related genes hypoxia-inducible factor 1α(HIF-1α)and Caspase3 mRNA were detected by real-time quantitative reverse transcription polymerase chain reaction(RT-qPCR).Results Compared with the normoxia group,after hypoxia induction,the cell viability of rat cardiomyocytes decreased significantly,the intracel-lular LDH activity and the levels of inflammatory factors IL-1β and IL-6 increased significantly,and the expression of Caspase3 and HIF-1α mRNA was significantly upregulated,with statistically significant differences(P<0.05 or P<0.01 or P<0.001);20 μmol/L SAL could significantly increase the viability of cardiomyocytes,reduce LDH ac-tivity and the levels of IL-1β and IL-6,and reduce the expression of Caspase3 and HIF-1α mRNA,with statistically significant differences(P<0.05 or P<0.01 or P<0.001).Conclusion SAL can effectively alleviate hypoxia-induced oxidative stress injury and inflammatory response in rat cardiomyocytes and inhibit cardiomyocyte apoptosis.关键词
