Abstract

Objective To explore the mechanism of action of RenShen Pingfei Fang(人参平肺方)in the treatment of pulmonary fibrosis(PF)using network pharmacology.Methods The active components of Renshen Pingfei Fang were obtained from the Traditional Chinese Medicine Systems Pharmacology Database and Analysis Platform(TCMSP),and the corresponding targets of the active components were retrieved from Swiss Target Predic-tion.The targets related to PF were screened from the Therapeutic Target Database(TTD)and DrugBank.The in-tersection of the targets of the active components and the disease-related targets was taken to obtain the potential tar-gets of Renshen Pingfei Fang against PF,and the target names were corrected using the UniProt database.The"drug-component-target"network was constructed by Cytoscape 3.9.0.The potential targets were subjected to Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway enrichment analysis.Results 50 intersection targets of drugs and diseases were screened out;the"drug-component-target"network identified 10 key active components such as quercetin,kaempferol,and β-sitosterol,and 14 core targets such as prostaglandin-en-doperoxide synthase 2(PTGS2),peroxisome proliferator-activated receptor gamma(PPARG),and β2-adrenergic re-ceptor(ADRB2);352 items were obtained from GO enrichment analysis;115 signaling pathways were obtained from KEGG pathway enrichment analysis,including phosphatidylinositol-3 kinase(PI3K)-protein kinase B(AKT),tumor necrosis factor(TNF),and mitogen-activated protein kinase(MAPK).Conclusion Renshen Pingfei Fang can exert anti-PF effects through multiple components,multiple targets,and multiple pathways.
