紫荚豌豆查尔酮异构酶PsCHI基因的克隆及表达分析
Cloning and Expression Analysis of Chalcone Isomerase PsCHI Gene in Purple Pea

Abstract
[Objective]The sequence of the CDS region of the purple pea CHI gene was cloned and subjected to bio-informatics analysis and tissue expression analysis to provide the theoretical basis for studying the molecular mechanism of CHI gene in pea anthocyanin synthesis.[Method]The sequence of the CDS region of the purple pea CHI gene(PsCHI)was cloned by PCR reaction,and analysis of the protein physicochemical properties and structure,the predication of the protein signal peptides,protein homology analysis,and the construction of phylogenetic trees were carried out by bio-informatics-related software,and then the relative expression levels of PsCHI genes were detected by real-time fluorescence quantitative PCR(qRT-PCR)technology.[Result]The CDS region of the purple pea CHI gene was 627 bp in length and encoded 209 amino acids.Purple pea PsCHI proteins were hydrophilic with no signal peptide,and the proteins were localized in the cytoplasm.The amino acid sequence had a conserved structural domain of the Chalcone 3 chalcone superfamily.The PsCHI gene had the highest relative expression level in purple pea pod tissues,which differed significantly(P<0.05)from the other tissues,and the relative expression level of the PsCHI gene in purple-pod pods was significantly(P<0.05)higher than that in green-pod pods.[Conclusion]The purple pea CHI gene(PsCHI)is characterized by a family of CHI genes and may have similar biological functions to CHI genes in other plants.The expression of CHI gene varies significantly in different tissues of pea and the highest relative expression level is found in purple pea pods,which is hypothesized to play an important role in the anthocyanin synthesis pathway in purple pea pods.关键词
