摘要

Abstract

[Objective]The key genes related to cellulose synthesis in purple chili fruits under two planting modes in the habitats of low latitude plateau were explored to provide a reference for studying the cellulose synthesis mechanism of purple chili fruits and selection of purple chili planting mode.[Method]The cellulose content of purple Capsicum annuum 16HN01 fruits was determined by spectrophotometry at six fruit development stages and the key genes related to cellulose synthesis and the planting mode were discovered by transcriptome sequencing,quantitative real-time fluorescence PCR(qRT-PCR)and correlation analysis.[Result]The cellulose content of purple C.annuum 16HN01 fruits in field and greenhouse modes gradually increased with fruit development process and was 3.79%-12.06% and 2.35%-9.55% respectively.The cellulose content of purple C.annuum 16HN01 fruits in greenhouse significantly decreased by 17.90%-38.09% compared with that in field.The transcriptional level of SGT,CesA2,CesA7,CSL-D3,CSL-G2,NAC,BES1 and MYB48 in purple C.annuum 16HN01 fruits planted in greenhouse was 17.13%,20.55%,17.73%,47.62%,33.68%,26.97%,42.00% and 29.00% lower than in field respectively.The transcriptional level of Korrigan gene(Kor)in purple C.annuum 16HN01 fruits planted in greenhouse was 64.94% higher than in field.The CC content of purple C.annuum 16HN01 fruits planted in field was significantly and positively related to CesA2,CSL-D3 and CSL-G2 and their correlation coefficients were 0.903,0.878 and 0.844 separately.The CC content of purple C.annuum 16HN01 fruits planted in greenhouse was positively related to CesA2 significantly and the correlation coefficients was 0.854.The CC content of purple C.annuum 16HN01 fruits planted in field was positively related to NAC and BES1 and their correlation coefficients were 0.560 and 0.413 respectively.The CC content of purple C.annuum 16HN01 fruits planted in greenhouse was positively related to NAC and the correlation coefficient was 0.671.[Conclusion]The structural gene(SG)and transcription factor gene(TF)related to cellulose synthesis of purple C.annuum fruits planted in field are mainly CesA2,CSL-D3 and CSL-G2,and NAC and BES1 in low latitude plateau respectively.The structural gene(SG)and transcription factor gene(TF)related to cellulose synthesis of purple C.annuum fruits planted in greenhouse are mainly CesA2,CSL-G2 and SGT,and NAC separately.The greenhouse planting mode can decrease cellulose content of purple C.annuum fruits and transcriptional level of the genes(except for Kor)related to cellulose synthesis.
