φ26,a lysogenic phage induced from the methicillin-resistant Staphylococcus aureus in Hainan

Abstract
Objective:To isolate and identify lysogenic phages in clinical isolates of methicillin-resistant Staphylococcus aureus(MRSA)from Hainan Island,determine the lytic ability of these phages,and explore their capacity for spreading virulence/resis-tance genes.Methods:Lysogenic phages were induced from MRSA clinical isolates using mitomycin C.By assessing their lytic ca-pabilities,a phage,designed as φ26,was subjected to the stability evaluation and the whole genome sequencing.Results:Isola-tion of lysogenic phages was evidenced in 53%of collected isolates(n=19).Among them,phage φ26 was featured as the highest lytic rate of 95.7%,which was demonstrated to be stable under conditions of pH 3-11 or temperatures below 60℃.Sequencing confirmed that its genome is a 42 061 bp linear double-stranded DNA(GC%=34.71%)no tRNA was detected,Including genes encoding leucocidin,a total of 66 open reading frames were identified.Conclusion:A novel lysogenic MRSA phage that may me-diate the horizontal transfer of clinical virulence genes was identified in this study,Whilst,its high lytic properties as well as prom-ise stabilities suggested the potential of its lysis associated elements for developing antibacterial agent development in future.关键词
