消痰化瘀利窍方抑制细胞焦亡减轻CIH小鼠胸主动脉损伤的研究
Study of Inhibitory Effect of Xiaotan Huayu Liqiao Formula on Cell Pyroptosis to Ameliorate Aortic Injury in CIH Mice

Abstract
Objective:To explore the inhibitory effect of Xiaotan Huayu Liqiao Formula(XHLF)on pyroptosis of thoracic aortic cells in mice with Chronic Intermittent Hypoxia(CIH)and its possible mechanism.Methods:Twenty-four healthy 8-week-old male C57BL/6N mice were randomly divided into a normoxic control group(Con group),a chronic intermittent hypoxia exposure group(CIH group),a chronic intermittent hypoxia+Xiaotan Huayu Liqiao Formula group(CIH+XHLF group),and a normoxic+Xiaotan Huayu Liqiao Formula group(Con+XHLF group).The CIH group and CIH+XHLF group were exposed to a CIH environment,and the Con+XHLF group and CIH+XHLF group were given Xiaotan Huayu Liqiao Formula by gavage daily,while the CIH group and Con group were given an equal volume of physiological saline,with a total intervention period of 28 days.After the modeling was completed,the blood pressure of the mice was monitored using the tail-cuff method;HE staining was used to observe the pathological changes in the thoracic aorta;the levels of endothelin-1(ET-1)and nitric oxide(NO)in the serum were detected;DHE staining was used to ob-serve the levels of reactive oxygen species(ROS),and colorimetric method to detect the content of malondialdehyde(MDA)and the activity of total superoxide dismutase(T-SOD)in the thoracic aorta;the positive cell rate of TUNEL staining in the thoracic aorta was observed;immunohistochemical staining was used to detect the protein expression of NOD-like receptor protein 3(NLRP3),Caspase-1,and interleukin-1β(IL-1β).Results:Compared with the Con group,the systolic blood pressure of the CIH group was significantly increased(P<0.05);the vascular intima was uneven,and the arrangement of cell nuclei was disordered;the content of ET-1 in-creased,and NO decreased(P<0.05);the levels of ROS and MDA increased;the activity of T-SOD decreased(P<0.05);the posi-tive cell rate of TUNEL staining in the aorta increased,and the protein expression of NLRP3,Caspase-1,and IL-1β was enhanced(P<0.05).Compared with the CIH group,the blood pressure of the CIH+XHLF group decreased(P<0.05);the vascular endothelial inju-ry was alleviated,ET-1 decreased,and NO increased(P<0.05);ROS and MDA decreased;T-SOD increased(P<0.05);the positive cell rate of TUNEL staining decreased,and the protein expression of NLRP3,Caspase-1,and IL-1β decreased(P<0.05).Conclusion:XHLF may ameliorate the thoracic aortic injury in CIH mice by inhibiting the ROS/NLRP3/Caspase-1 pathway.关键词
