摘要

Abstract

Objective:To use fecal microbiota transplantation(FMT)technology to verify the mechanism of Danggui Shaoyao San in regulating lipid metabolism and protecting the liver.Methods:Fifty SD rats were divided into a control group,a metabolic-associated fatty liver disease(MAFLD)group,and low,medium,and high dose groups of Danggui Shaoyao San.Except the control group,the other groups were fed with high-fat feed for 8 weeks to establish the MAFLD model,and each dosage group of Danggui Shaoyao San was trea-ted the corresponding does of the drug by intragastric administration for 4 weeks.After treatment,feces were collected to prepare fecal bacterial solution.Another 60 SD rats were randomly divided into a control group,an MAFLD rat fecal microbiota transplantation group(MO-FMT group),a normal rat fecal microbiota transplantation group(C-FMT group),and low,medium,and high dose Danggui Shaoyao San rat FMT groups.Except for the control group,the other groups were given antibiotic-free water for 4 weeks to establish a pseudo-germfree rat model.After the model was successfully established,the rats in each group were given corresponding fecal bacterial solution intervention for 4 weeks.Serum,liver tissue,ileum and colon tissues,and feces were collected from the rats in each group to de-tect related indicators.Results:Compared with the control group,the levels of serum total cholesterol(TC),triglycerides(TG),low-density lipoprotein(LDL-C),alanine aminotransferase(ALT),aspartate aminotransferase(AST),tumor necrosis factor-α(TNF-α),as well as the content or activity of TC,TG,and free fatty acids(FFA)in the liver tissue in the MO-FMT group were all increased(P<0.05).A small number of inflammatory cell infiltrations were visible in the hepatocytes.The content of serum high-density lipoprotein(HDL-C)and interleukin-10(IL-10),the α-diversity index,the abundance of Bacteroidetes,and the protein expression levels of occlu-din and ZO-1 in the intestine were all decreased(P<0.05),while the abundance of Firmicutes,the F/B value,and the protein expres-sion levels of Toll-like receptor 4(TLR4)and phosphorylated nuclear factor-κB(p-NF-κB)in the liver tissue were all increased(P<0.05).Compared with the MO-FMT group,the hepatocytes in each FMT group of Danggui Shaoyao San showed no obvious damage,and both liver function and lipid metabolism were tending towards normal(P<0.05),up-regulate the content of serum IL-10,the α-diversity index,the abundance of Bacteroidetes,and the protein expression levels of occludin and ZO-1(P<0.05),and down-regulate the content of serum TNF-α,the abundance of Firmicutes,the F/B value,and the protein expression levels of TLR4 and p-NF-κB in the liver tissue(P<0.05).Conclusion:Danggui Shaoyao San exerts its lipid-regulating and liver-protecting effects by reshaping the abundance of in-testinal microbiota,improving the function of intestinal mucosal mechanical barrier,and inhibiting the TLR4 inflammatory pathway.
