精子相关抗原5在肝内胆管癌中表达的临床意义及其对癌细胞增殖、迁移和侵袭的影响
Clinical significance of sperm-associated antigen 5 expression in intrahep-atic cholangiocarcinoma and its effects on cancer cell proliferation,mi-gration,and invasion

Abstract
Objective To detect the expression of sperm-associated antigen 5(SPAG5)in intrahepatic cholangiocarcinoma(ICC),the clinical significance was analyzed,and its impact on cancer cell proliferation,migration,and invasion were clarified.Methods The 6 paired fresh and 56 paraffin embedded ICC patients'cancer tissues and adjacent normal tissues treated at Dongguan Waterfront Zone Central Hospital,Guangdong Province from January 2016 to December 2020 were collected and were as the research subjects,the expression of SPAG5 in fresh and paraffin embedded ICC tissues and ICC cell lines was detected by quantitative real-time polymerase chain reaction,western blot,and immunohistochemistry,respectively,the correlation between its expression and the clinical pathological characteristics and prognosis of ICC patients were analyzed.The 5-ethynyl-2'-deoxyuridine test,clone formation assay,and Transwell chamber assay were used to detect the changes in proliferation,clone formation,migration,and invasion ability of ICC cells after overexpression or knockdown of SPAG5.Results The expression of SPAG5 mRNA and protein in ICC cancer tissues were higher than those in adjacent normal tissues,and the differences were statistically significant(P＜0.05).Patients with high expression of SPAG5 had shorter tumor free survival and overall survival than those with low expression of SPAG5(P＜0.05).Overexpression of SPAG5 significantly upregulated the proliferation,migration,and invasion abilities of ICC cell RBE and HCC C9810,while knockdown of SPAG5 expression significantly downregulated the proliferation,migration,and invasion abilities of ICC cell QBC939(P＜0.05).Conclusion The high expression of SPAG 5 in ICC predicts poor prognosis in ICC patients and enhances the proliferation,migration and invasion of ICC.关键词
