表达传染性喉气管炎病毒gB蛋白重组耐热新城疫病毒的构建
Generation of the thermostable recombinant Newcastle disease virus expressing the gB protein of infectious laryngotracheitis virus

Abstract
To generate a thermostable and combined vaccine candidate against the infectious laryngotracheitis(ILT)and Newcastle disease(ND),this study cloned the gB gene of ILTV to the thermostable NDV NDV/rAHR09 strain using the seamless cloning technique.Then,the Western blot experiment was used to check the expression of gB,and the thermostable and virulence assessment experiments were used to detect the biological characteristics of the recombinant vi-rus.The results showed that the gB gene of ILTV was successfully inserted into the pNDV/rAHR09,and the rescued recombinant virus showed a high haemagglutination(HA)titer.Additionally,the Western blot assay confirmed the ex-pression of the gB protein.The thermostable assessment showed that the recombinant viruses maintained a high HA ac-tivity and virus titer after exposure to 56 ℃ for about 60 minutes.Pathogenicity assessment data indicated that the recom-binant virus had a mean death time(MDT)in chicken embryos of 148 hours,an intracerebral pathogenicity index(ICPI)of 0,and an intravenous pathogenicity index(IVPI)of 0.1,suggesting that the recombinant virus possessed typical char-acteristics of an attenuated NDV.In conclusion,the thermostable vaccine candidate for ILT and ND was successfully de-veloped in this study,which provides a novel approach for developing a live vector vaccine against ILTV.关键词
