Abstract

In order to prepare a monoclonal antibody against the E1 protein of Getah virus(GETV),the hydrophilic region fragment(amino acid 31-182 region)of GETV E1 gene was constructed into pCold-TF prokaryotic expression vector in this study.After expression and purification,TF-E1 recombinant protein was prepared,and mice were immu-nized with it as an immunogen.With the help of hybridoma technology,one monoclonal antibody against GETV E1 pro-tein was successfully obtained.The biological characterization of the obtained E1 monoclonal antibody showed that the monoclonal antibody against E1 protein prepared in this study was suitable for indirect immunofluorescence and Western blot characterization.Meanwhile,by truncating the expression of E1 protein,the antigenic epitope recognized by the E1 protein monoclonal antibody was further clarified to be 73RPDYQC78,and the comparison of this antigenic epitope with the sequence of GETV strains that had been uploaded in GenBank revealed that this sequence was highly conserved among GETV strains.In conclusion,the successful preparation of monoclonal antibody to GETV E1 protein provides an immu-nological tool for the study of GETV.
