CACNA1C和RYR2基因核酸多态性与蛋壳品质的关系
Relationship between polymorphism of CACNA1C and RYR2 gene promoter region and eggshell quality

Abstract
The up-regulation of CACNA1C(CaV1)expression in the calcium regulation pathway inhibits the expres-sion of ryanodine receptor(RYR2)gene and promotes calcium ion transport and eggshell formation.This study analyzed the polymorphism of CACNA1C and RYR2 gene promoter region and its relationship with eggshell quality of Wuhua chicken.Total DNA was extracted from wing vein blood of 340 35-week-old Wuhua chickens with complete egg produc-tion records.PCR amplification and sequencing were used to screen SNPs in the promoter regions of CACNA1C and RYR2 genes.Eggs were collected for 3 days for eggshell quality detection,and 5 chickens were randomly slaughtered to take heart,liver,spleen,lung,kidney,chest muscle,leg muscle,ovary,enlarged part,uterus,isthmus and other tis-sues for gene expression quantification.The overexpression and knockdown plasmids of CACNA1C gene were constructed and transfected into DF-1 cells to analyze their regulatory effects on the expression of RYR2 gene.The results showed that one SNP site was screened at-2 000 bp in the promoter region of CACNA1C and RYR2 genes,respectively.CACNA1C gene g.61191372 C>G locus produced three genotypes.The eggshell strength of CC was significantly higher than that of CG(P<0.05).The g.29842706 G>A locus of RYR2 gene produced three genotypes.The eggshell thick-ness of AG and AA was significantly higher than that of GG(P>0.05).The expression of CACNA1C in the enlarged part was significantly higher than that in the uterus,ovary,isthmus,heart,liver,spleen,lung,kidney,chest muscle and leg muscle.Overexpression of CACNA1C gene CC type inhibited RYR2 expression.In conclusion,the CC type of CACNA1C gene promoter region can reduce the expression of RXR2 gene,promote calcium ion transport,increase egg-shell density,and improve eggshell strength.关键词
