超声处理对泡核桃雄花总多酚提取效果的影响及其特性
Effects of ultrasound treatment on the extraction of Juglans sigillata Dode male flower total polyphenols and its characteristics

Abstract
The Juglans sigillata Dode male flower extracts can reduce the oxidative stress and inhibit the oxidation of macromolecules.In this study,on the basis of optimizing the better extraction conditions of male flower total polyphe-nols,its components and antioxidant effect in vitro were investigated,and the ultrasound mechanism was preliminarily described.Total polyphenols extraction conditions of male flowers was optimized by multiple quadratic regression equa-tion;its components were analyzed by HPLC;The antioxidant activity of total polyphenol was evaluated by determining the clearance of superoxide anion and hydroxyl radical;The microstructure of walnut male flowers powders before and af-ter ultrasound treatment was observed by SEM.The results showed that the ultrasound power was determined at 250 W,the better extraction process of total polyphenol of walnut male flowers was as follows:solid to liquid ratio 1:60,treat-ment time 21 min,ethanol concentration 50%,treatment temperature 61 ℃,the total polyphenol yield of walnut male flowers was(24.45±2.12)mg·g-1;Four kinds of monomeric phenol were identified by HPLC from walnut male flow-ers.Among them,chlorogenic acid and rutinum were higher,and gallic acid and quercetin were lower;total polyphenol,vitamin C(Vc)and BHT could remove superoxide anions and hydroxyl radicals.In terms of its clearance capability,Vc>total polyphenol>BHT;ultrasound treatment could tear and destroy the tissue surface of male flower,and release more plant polyphenol and strengthen the extraction effect.The results can provide the possibility for its application of male flowers polyphenols in the field of food storage and preservation.关键词
