黄腐酚对口腔鳞癌细胞增殖能力的影响
Effect of xanthohumol on the proliferation ability of oral squamous cell carcinoma cells

Abstract
Objective:To investigate the effect of xanthohumol on the proliferation and apoptosis of human oral squa-mous cell carcinoma cells SCC-9 and CAL-27.Methods:SCC-9 and CAL-27 cells were treated with different concentrations(0,0.1,0.2,0.5,1,2,5,10,20 μmol/L)of xanthohumol for 24 h,48 h and 72 h.The effect of xanthohumol on the prolifera-tion of SCC-9 and CAL-27 cells was detected by CCK-8 method,and the subsequent experimental concentrations(0,2,5,10 μmol/L)were selected according to the IC50 value.EdU was used to detect the effect of xanthohumol on the proliferation of SCC-9 and CAL-27 cells.The effect of xanthohumol on the apoptosis of SCC-9 and CAL-27 cells was detected by flow cytom-etry.The expression levels of Skp2 and other proteins treated with xanthohumol for 48 h were detected by Western Blot.The mRNA expression of Skp2 in cells was detected by qRT-PCR.Results:Within a certain concentration range,xanthohumol(2,5,10 μmol/L)could inhibit the proliferation of oral squamous cell carcinoma SCC-9 and CAL-27 cells in a time-dependent and dose-dependent manner(P<0.05).At the same time,after the intervention of xanthohumol(0,2,5,10 μmol/L)for 48 hours can promote the apoptosis of SCC-9 and CAL-27 cells(P<0.001),and down-regulate the expression levels of Skp2 in SCC-9 and CAL-27 cells,and the mRNA expression of Skp2 decreased with the concentration(P<0.001).Conclusion:A certain concentration of xanthohumol can effectively inhibit the proliferation of oral squamous cell carcinoma cells and induce the apoptosis of oral squamous cell carcinoma cells.The mechanism may be related to the down-regulation of the expression of cell cycle protein Skp2 by xanthohumol.关键词
