Abstract

The purpose of this study was to explore the effects of hypotaurine and heparin on the incubation effect of sika deer sperm in vitro.Modified Tyrode's solution(TALP),synthetic oviduct fluid(SOF),and modified synthetic oviduct fluid(SOF2)containing 0,10,20,and 40 mg/L of hypotaurine were used to incubate sika deer sperm in vitro.The sperm of sika deer were incubated in vitro with TALP and SOF containing 0,5,20 mg/L heparin and 20 mg/L hypotaurine.Finally,sika deer oocytes were fertilized in vitro with SOF-20%sheep serum(SS)and SOF-5 mg/L heparin.The sperm motility rate at 18 h in vitro incubation,linear motility sperm rate and embryo development after in vitro fertilization were measured.The results showed that at 18 h incubation,the sperm motility rate and linear motility rate of all semen increased with the increase of the concentration of hypotaurine,and the highest was reached when the concentration of hypotaurine was 20 mg/L.Heparin can decrease the sperm motility rate and increase the linear motility sperm rate of sika deer.When the concentration of heparin reaches 20 mg/L,the increase trend of linear motility sperm rate begins to decline.The fertilized deer oocytes were fertilized in in vitro with SOF-20%SS,and the fertilized eggs could develop to the morula stage at the highest.In vitro fertilization with SOF-5 mg/L heparin,the fertilized egg only develops to the 4-cell stage at most.The results showed that hypotaurine was beneficial to the in vitro incubation of sika deer sperm,and the optimal concentration was 20 mg/L.Heparin is not conducive to the in vitro incubation of sika deer sperm,and its in vitro fertilization effect is far less than 20%SS,and the synergistic effect with SS is not obvious.
