Abstract

The study aimed to optimize the prescription and preparation process of cefotiam hydrochloride suspension through single-factor tests and response surface methodology.The effects of the amounts of suspending agent,wetting agent,and antioxidant on the comprehensive score(Y value)of the suspension were investigated.Based on this,the Box-Behnken response surface methodology was used,selecting the amounts of suspending agent,wetting agent,and antioxidant as three factors,with the Y value as the response value.The optimal preparation process conditions for cefotiam hydrochloride suspension were determined through response surface experiments.The results showed that the optimal preparation process parameters for cefotiam hydrochloride suspension were a suspending agent amount of 3.5%,a wetting agent amount of 2.13%,and an antioxidant amount of 0.5%.Three parallel validation tests were conducted under optimal conditions,and the average value obtained for the Y value of cefotiam hydrochloride suspension was 1.21%.The study demonstrated that the method for preparing cefotiam hydrochloride suspension optimized by single-factor tests and response surface methodology was feasible.
