Abstract

The study aimed to develop microsatellite markers in the sheep genome to provide effective molecular markers for the conservation of sheep breeds and genetic research.Through next-generation sequencing,24 microsatellite markers were developed for four indigenous Chinese sheep breeds(Lanzhou fat-tailed sheep,Tan sheep,Tibetan sheep,and small-tailed han sheep).These 24 markers were used to screen 11 Lanzhou fat-tailed sheep,19 Tan sheep,30 Tibetan sheep,and 12 small-tailed han sheep collected from Gansu Province.The results showed that in the four sheep breeds,the number of alleles per locus ranged from 2 to 11(with an average of 4).After Bonferroni correction,several loci significantly deviated from Hardy-Weinberg equilibrium(2 in Lanzhou fat-tailed sheep,9 in Tan sheep,7 in Tibetan sheep,and 2 in Xsmall-tailed han sheep),which may be the result of non-random mating.The study demonstrated that the developed markers were accurate and effective,with high polymorphism,and could be used for genetic research and genetic management of indigenous Chinese sheep breeds.
