基于NGS技术的4种绵羊品种微卫星标记开发与鉴定
Development and identification of microsatellite loci in four sheep breeds based on NGS technology

Abstract
The study aimed to develop microsatellite markers in the sheep genome to provide effective molecular markers for the conservation of sheep breeds and genetic research.Through next-generation sequencing,24 microsatellite markers were developed for four indigenous Chinese sheep breeds(Lanzhou fat-tailed sheep,Tan sheep,Tibetan sheep,and small-tailed han sheep).These 24 markers were used to screen 11 Lanzhou fat-tailed sheep,19 Tan sheep,30 Tibetan sheep,and 12 small-tailed han sheep collected from Gansu Province.The results showed that in the four sheep breeds,the number of alleles per locus ranged from 2 to 11(with an average of 4).After Bonferroni correction,several loci significantly deviated from Hardy-Weinberg equilibrium(2 in Lanzhou fat-tailed sheep,9 in Tan sheep,7 in Tibetan sheep,and 2 in Xsmall-tailed han sheep),which may be the result of non-random mating.The study demonstrated that the developed markers were accurate and effective,with high polymorphism,and could be used for genetic research and genetic management of indigenous Chinese sheep breeds.关键词
