Abstract

The experiment aims to develop an identification method for Streptococcus danieliae and to study its drug resistance and susceptibility characteristics.The experiment first involved the isolation and culture of the standard strain of Streptococcus danieliae CCUG57647(Sd57647),Gram staining and microscopic examination,biochemical identification,and 16S rRNA sequence analysis to determine its characteristics and establish detection methods.This was followed by verification of the detection method using five isolates separated from the tracheal secretions of SPF mice.One of the isolates Sd001,was observed for its morphological characteristics under a transmission electron microscope and subjected to a salt tolerance test.Subsequently,it was cultured up to the 20th generation on Columbia blood agar plates to examine its growth stability.The results showed that Streptococcus danieliae is a Gram-positive streptococcus with a visible capsule structure under the transmission electron microscope.On Columbia blood agar,it forms circular,smooth,semi-transparent,pale green colonies with α-hemolysis(1~2 mm in diameter)after cultivation at 36℃for 24~48 hours.Fifteen biochemical reactions with specific identification characteristics for Streptococcus danieliae were selected by comparing the biochemical identification results with those of Streptococcus thoraltensi(St001),and the results of these 15 biochemical identification reactions in the isolates from the tracheal secretions of SPF mice were consistent with those of Sd57647.The 16S rRNA phylogenetic tree analysis showed that the isolates clustered with Streptococcus danieliae.After 20 generations of subculturing,the isolates maintained stable morphological structures and biochemical reactions,but their drug susceptibility changed.The study established a detection method for Streptococcus danieliae and preliminarily investigated its phenotypic characteristics,laying the foundation for the quality control of laboratory animals.
