Abstract

This study aims to optimize the extraction process of total alkaloids from Peganum nigellastrum Bunge,taking the extraction rate of total alkaloids from Peganum nigellastrum Bunge as the evaluation index,the methods of cation exchange resin extraction,acid-solution and alkali-isolation extraction,and organic solvent extraction were compared,and the optimal extraction method was determined to be acid-solution and alkali-isolation extraction.Single factor experiments combined with response surface method were used to optimize the acid-solution and alkali-isolation extraction process.In the single factor experiment,factors such as powder particle size,soaking time,hydrochloric acid concentration,and hydrochloric acid dosage were investigated.With the optimal value of each single factor as the central point,Box-Behnken's central combination method was used to design the factors and levels of the response surface,and optimize its extraction process conditions.The experimental results showed that when the powder particle size,soaking time,HCl concentration and HCl dosage were 60 mesh,49 h,5%and 40 mL,respectively,the extraction effect of total alkaloids from Peganum nigellastrum Bunge was the best,with a yield of 32.91%,which was close to the predicted value of the model(33.082%).Through this experimental study,a quadratic model equation was obtained that could better predict the experimental results,which laid a preliminary foundation for subsequent research on the pharmacological effects of total alkaloids from Peganum nigellastrum Bunge.
