Abstract

This study investigates the dynamic changes in starch accumulation,sucrose accumulation,activities of starch synthesis-related enzymes,and relative expression levels of key genes during the grain filling period of four barley varieties(Mengpi 3,Ganpi 4,Mengpi 5,and Kenpi 7)under varying nitrogen fertilizer rates(0,90,180 and 270 kg/hm2 pure nitrogen),planting densities(3.75,4.50,5.25 and 6.00 million plants/hm2),and soil water con-tents(20%,40%,60%and 80%of field maximum water holding capacity).Through systematic analysis of correla-tions between starch accumulation,sucrose accumulation,and the relative expression levels of HvAGPase,HvStS and HvSBE,a molecular biology-based model of starch formation in barley grains was constructed.The model focuses on the relative expression levels of genes encoding key enzymes regulating starch formation,namely ADP-glucose pyro-phosphorylase polypeptide(AGPase),starch synthase(StS),and starch branching enzyme(SBE).Model validation re-sults showed an absolute prediction error of 5.13-18.99 g/m2,with root mean square error(RMSE)of 10.421 4 g/m2,relative root mean square error(RRMSE)of 0.431 2,model efficiency(EF)of-0.004 6,and residual coefficient(CRM)of 0.998 1,indicating good mechanistic accuracy and predictive capability.
