血浆miRNA-28-3P和EBV-DNA检测对鼻咽癌患者的诊断价值研究
Diagnostic Value of Plasma miRNA-28-3P and EBV-DNA in Patients with Nasopharyngeal Carcinoma

Abstract
Objective:To evaluate the diagnostic value of plasma micro ribonucleic acid(miRNA)-28-3P and epstein-barr virus deoxyribonucleic acid(EBV-DNA)in patients with nasopharyngeal carcinoma.Methods:The medical records of 52 newly treated nasopharyngeal carcinoma patients admitted to Ganzhou Cancer Hospital from January 2021 to August 2024 were retrospectively analyzed and included in the study group,and 48 healthy people in the same period with matching gender and age were analyzed and included in the control group.Real-time fluorescence quantitative polymerase chain reaction was used to determine the expression levels of miRNA-28-3P and EBV-DNA in the two groups.The value of miRNA-28-3P and EBV-DNA in the diagnosis of nasopharyngeal carcinoma was analyzed by receiver operating characteristic.Flow cytometry was used to detect the expression level of CD4+/CD8+in peripheral blood of the two groups,and the correlation between miRNA-28-3P,EBV-DNA and CD4+/CD8+was analyzed.Results:The plasma miRNA-28-3P level of the study group was lower than that of the control group,and the plasma EBV-DNA level was higher than that of the control group,the differences were statistically significant(P<0.05).The sensitivity and specificity of miRNA-28-3P in the diagnosis of nasopharyngeal carcinoma were 89.1%and 65.2%respectively;the sensitivity and specificity of EBV-DNA in the diagnosis of nasopharyngeal carcinoma were 50.5%and 93.2%respectively;the sensitivity and specificity of the combined diagnosis of nasopharyngeal carcinoma were 85%and 85%.miRNA-28-3P was positively correlated with CD4+/CD8+(r=0.621,P<0.001),and EBV-DNA was negatively correlated with CD4+/CD8+(r=-0.701,P<0.001).Conclusion:The abnormal expression of miRNA-28-3P and EBV-DNA in the plasma of patients with nasopharyngeal carcinoma is related to CD4+/CD8+in peripheral blood of patients,and the combined detection of miRNA-28-3P and EBV-DNA is expected to be a potential biomarker for diagnosis and treatment and prognosis of nasopharyngeal carcinoma.关键词
