Abstract

Objective:To study the mechanism of action of Jianaikang condensed pill in the treatment of AIDS based on net-work pharmacology and clinical trials.Methods:The main active ingredients and targets of Jianaikang condensed pill were screened by TCMSP,ETCM,PubChem and Swiss Target Prediction databases,and the disease targets were obtained from Gen-eCards,OMIM,TTD and DrugBank databases,and the Venny 2.1.0 online tool was used to draw the Wayne diagrams of the drug and the disease targets.Using Venny 2.1.0 online tool to draw the Wayne diagram of drug and disease target,obtain the in-tersection target,and carry out protein interaction analysis with the help of STRING database;using Cytpscape 3.10.1 software to construct the network diagram of drug-active ingredient-intersection target and protein interaction network;and carrying out GO and KEGG functional enrichment analysis of core effector targets in David database.Serum IL-17,TNF-α and IFN-γ counts were detected before and after treatment in patients in the AIDS Jianaikan group and the control group.Results:After preliminary screening,90active ingredients and 509potential targets such as quercetin,kaempferol,Gomisin B,and Celabenzine were ob-tained,among which 172targets intersected with AIDS,and the core targets included TP53,TNF,AKT1,ACTB,IL-6,CASP3,IL-1B,BCL-2,EGFR,and NF-κB1,etc;GO was enriched in BP positive regulation of gene expression,negative regulation of apoptotic process,promotion of inflammatory response,etc;MF mainly includes ATP-binding,enzyme-binding,protein-binding,etc.CC mainly includes extracellular space,chromatin,cytosolm,etc;KEGG-enriched discovery of signal-ling pathways,such as IL-17,PI3K/AKT,and TNF.In the clinical trial,compared with the pre-treatment,the counts of IL-17,TNF-α and IFN-γ in the Jian Aikang group were significantly higher after treatment(P<0.05),while the indicators in the control group fluctuated to some extent,but there was no significant difference(P>0.05);the counts of the indicators in the two groups before and after the treatment did not differ from those of the patients(P>0.05).Conclusion:Jianaikang condensed pill can improve the levels of cytokines IL-17and IFN-γ to a certain extent,and promote the improvement of immune function,in which the mechanism that plays a major role may be the regulation of IL-17pathway and PI3K/Akt pathway.
