Experimental study on the effects of Tongfeng Jinhuang gel plaster on knee swelling,uric acid levels,and inflam-matory factors in rats with acute gouty arthritis

Abstract
Objective:This study aimed to assess the therapeutic effects of Tongfeng Jinhuang gel plaster on acute gouty ar-thritis(AGA)in a rat model.Methods:Sixty Sprague-Dawley rats were randomly assigned to six groups:blank group,model group,colchicine group(0.3mg·kg-1),colchicine+low-dose gel plaster group(TFJH-L,0.3mg·kg-1+72mg·kg-1),colchicine+medium dose gel plaster group(TFJH-M,0.3mg·kg-1+144mg·kg-1),colchicine+high-dose gel plaster group(TFJH-H,0.3mg·kg-1+288mg·kg-1),with 10rats in each group.The blank group and model group were administered an equivalent volume of distilled water via gavage for a period of seven consecutive days.On the fifth day of the experiment,rats in all groups(excluding the control group)were injected with0.2ml of monosodium urate(MSU)solution(25g·L-1)into the right knee joint cavity to induce acute gouty arthritis.The control group received an equivalent volume of sterile physiological sa-line injection.Following the induction of the model,medication was administered for an additional two days,and on the seventh day,synovial tissue from the knee joint was extracted to assess the extent of swelling in the rat knee joint.The serum levels of u-ric acid(SUA),C-reactive protein(CRP),tumor necrosis factor α(TNF α),interleukin-1β(IL-1β),interleukin-18(IL-18),and interleukin-6(IL-6)were quantified,and the knee joint pathology was assessed.Results:The model group of rats exhibited significant knee joint swelling and elevated levels of SUA,CRP,TNF-α,IL-1β,IL-18,and IL-6compared to the control group(P<0.01).Fibrous tissue proliferation and structural abnormalities were noted in the synovial tissue of the knee joint,accompanied by a substantial presence of inflammatory cells and blood vessel congestion.In comparison to the control group,the Colchicine group,TFJH-L group,TFJH-M group,and TFJH-H group exhibited significantly decreased joint swell-ing,as well as reduced levels of serum SUA,CRP,TNF-α,IL-1β,IL-18,and IL-6(P<0.05).The administration of Colchicine resulted in a reduction in synovial tissue proliferation,inflammatory infiltration,and vascular congestion in the knee joint.Similarly,the TFJH-L,TFJH-M,and TFJH-H groups exhibited a notable decrease in synovial tissue proliferation,in-flammatory infiltration,and relief of vascular congestion in the knee joint.Conclusion:Tongfeng Jinhuang gel plaster demon-strates efficacy in reducing knee joint swelling,lowering inflammatory factor levels,ameliorating synovial hyperplasia,and de-creasing inflammatory cell infiltration in the knee joint.This suggests that it may offer a novel therapeutic option for the manage-ment of acute gouty arthritis in clinical practice.关键词
