构树根萌条水培生根发育及解剖观察
Hydroponic Rooting Development and Anatomical Observation of the Root Sprout of Broussonetia papyrifera

Abstract
Broussonetia papyrifera has important ecological and economic value,can not only be used as an excellent pioneer tree to protect the environment and repair soil pollution,but also used as a high-quality source of livestock feed,providing protein and energy for animals.In this paper,the root sprouts of B.papyrifera of Guangzhou germplasm were used as experimental materials,and orthogonal tests of four factors and four levels of NAA concentration,IBA concentration,Hoagland concentration and sprout maturity were designed to determine the optimal hydroponic treatment method.At the same time,paraffin sections were taken from the root sites of the sprouts with different hydroponic rooting times,to observe the process of adventitious root formation.The results showed that the effects of NAA,IBA,Hoagland concentration and maturity on rooting effect index were very significant.Among them,the optimal treatment level of NAA is 0.1 mg/L,the optimal treatment level of IBA is 0 mg/L,the optimal treatment level of Hoagland concentration is 1/4,and the optimal maturity level is 3(retaining 6 leaves).According to the cross-cutting observation of the root sprout base in different hydroponic periods,the rooting process of the sprout can be divided into three periods,among which the formation period of root primordium on day 5~6,the occurrence period of adventitious roots on day 9~12,and after 12 d is the period of adventitious root elongation.Adventitious roots started to appear around 9 d of hydroponic cultivation of the root sprouting sprouts of B.papyrifera,and the adventitious root primordia originated from the formation layer,which was the induced root primordia.Hydroponic rooting was best when using 1/4 Hoagland+NAA 0.1 mg/L and maturity 3(retaining 6 leaves)root sprouting spike strips of conifers,with 100%rooting rate and rooting effect index of 42.55.关键词
