Calcineurin inhibitor tacrolimus interferes CVB3-induced autophagy

Abstract
Objective To investigate the role of tacrolimus,a calmodulin phosphatase inhibitor,on coxsackievirus B3-induced cellular autophagy.Methods The cells were divided into control group,CVB3-infected group,and CVB3-infected+tacrolimus group,and the expression levels of LC3Ⅰ,LC3Ⅱ,p62,and VP1 proteins were detected by Western blotting,respectively.RT-qPCR was used to detect the adsorption of tacrolimus on CVB3 viral genome.The expression of tacrolimus on the titer of CVB3 virus was judged according to the half tissue infectious dose(TCID50).Results CVB3 infection induced an increase in the expression level of autophagy,and CVB3-induced autophagy showed a decreasing trend with the addition of tacrolimus.Tacrolimus down-regulated the expression of CVB3 viral capsid protein VP1 and decreased the adsorption of CVB3 viral genome and viral titer.Conclusion Tacrolimus reduces CVB3 viral gene replication and improves CVB3-induced cellular autophagy.关键词
