摘要

Abstract

Objective To detect and analyze the quantity,diversity,abundance,and aflatoxins(AFs)production characteristics of contaminated mold in 16 plant types of Chinese materia medica decoction pieces(CMMDPs)that are susceptible to aflatoxin contamination,and to explore the symbiotic fungal community characteristics of Aspergillus flavus which produce aflatoxins.Methods The number of contaminated mold in samples was detected using the coating culture method,and typical colonies of different mold were counted,isolated,and purified and then the purified strains were identified using fungal ITS sequencing combined with microscopic characteristics,and a phylogenetic tree was plotted.After isolated Aspergillus flavus were cultured for aflatoxins production,AFs were detected using immunoaffinity column purification HPLC post column photochemical derivatization method.Results Out of 43 batches of samples,11 batches had a contaminated mold count of less than 10 cfu·g-1,while the rest of the samples ranged from 6.8×101~6.9×103 cfu·g-1.After identification,the contaminated molds belonged to 7 genera and 16 species,and 8 batches of samples had more than 3 types of contaminated molds.Six strains of Aspergillus flavus were isolated,of which four were aflatoxins producing strains with different aflatoxins producing abilities.The total number of contaminated mold in the samples from the four strains is generally greater than 103 cfu·g-1,with more than three species and a rich population structure.Most of them have close genetic relationships,which exhibits certain colony characteristics.Conclusion There is a certain correlation between the microbial community structure of contaminated mold in CMMDPs and the growth,aflatoxin production ability of Aspergillus flavus,which can be used for early pollution warning,biological control,etc.,providing new ideas for the prevention and control of aflatoxin contamination in CMMDPs.
