柱后衍生阳离子交换色谱法测定依降钙素中氨基酸组成与肽含量
Determination of amino acid composition and peptide content in elcatonin by post-column derivatization cation-exchange chromatography

Abstract
Objective To determine the amino acid composition and peptide content in elcatonin using amino acid analyzer and compare the results with those obtained from the pre-column derivatization high-performance liquid chromatography(HPLC)method.Methods The chromatographic column was 2622SC-PH(4.6 mm×60 mm).The flow rate of pump A(pH series solution)was 0.4 mL·min-1,and the flow rate of pump B(ninhydrin solution)was 0.35 mL·min-1.The detection wavelengths were set at 440 nm(proline)and 570 nm(other amino acids).Results The results showed that the linear relationship of the 14 amino acids was good,the repeatability RSD was 1.1%~2.5%,and the recovery was 98.0%-103.6%.The results were generally consistent with those obtained from the HPLC method.Conclusion The amino acid analyzer method was simple and highly automated,and can be used to determine the amino acid composition and peptide content in elcatonin.关键词
