Abstract

The experiment aims to investigate the inhibitory effects of Areca catechu extract(ANE)and its four main alkaloids(arecoline,arecaidine,guvacine,and guvacoline)on jack bean urease(JBU),using acetoxyhydroxamic acid(AHA)as a control,and to further explore the sites of their inhibitory action.The results showed that guvacoline(Guv)exhibited the most significant JBU inhibition effect with an IC50 value of 31.06 mg/L.Urease kinetics experiments showed that both ANE and Guv are hybrid inhibitors of JBU.The inhibitory effects of ANE and Guv on JBU were significantly reduced in the presence of thiol reagents(P<0.05),and the inhibitory effects of Guv on JBU were significantly elevated in the presence of Ni2+competitive inhibitors(P<0.05).Molecular docking results showed that Guv formed interactions with amino acid residues Asp 494,Ala 440,His 519,His 593,Arg 609,Gly 550,His 492,Ala 636,Asp 633,and His 409.The study indicates that Guv can effectively reduce the catabolism of urea by JBU,and Areca catechu possesses the potential for novel urease inhibitor functional product development.
