磨盘草多糖的提取工艺优化、结构表征及其抗氧化活性
Optimization of extraction process,structural characterization,and antioxidant activity of polysaccharide from Abutilon indicum(L.)Sweet

Abstract
The extraction rate of Abutilon indicum(L.)Sweet polysaccharide was used as the evaluation index.The water extraction process of Abutilon indicum(L.)Sweet polysaccharide was optimized by single-factor experiments and response surface methodology.The structure of the polysaccharide was characterized by ultraviolet spectroscopy(UV),Fourier transform infrared spectroscopy(FT-IR),particle size and Zeta potential analyzer,and scanning electron microscopy(SEM).The in vitro antioxidant activity of the polysaccharide was also evaluated.The results showed that under the conditions of liquid-to-material ratio of 43 mL/g,extraction temperature of 80℃,and extraction time of 167 min,the extraction rate of the polysaccharide reached 11.26%.The content of proteins and nucleic acids in Abutilon indicum(L.)Sweet polysaccharide was low,and characteristic absorption signals of polysaccharide was present.The average particle size was 493.11 nm,and the average Zeta potential was-22.78 mV.The microstructure was characterized by smooth short rod-like structures with evenly distributed and closely arranged sponge-like pores.It had a certain scavenging ability on 1,1-diphenyl-2-trinitrophenylhydrazine(DPPH)and 2,2-diazo-di(3-ethyl-benzothiazolin-6-sulfonic acid)diamiammonium salt(ABTS)radicals,with EC50 values of 4.99 g/L and 0.28 g/L,respectively.The study indicates that the experimental results can provide a reference for the industrial extraction and activity research of Abutilon indicum(L.)Sweet polysaccharide,and promote its wide application as feed additive in animal production.关键词
