摘要

Abstract

Objective To investigate the chemical constituents and in vitro antitumor activities of secondary metabolites derived from the mangrove-associated Actinobacteria Kribbella karoonensis A01098.Methods The liquid fermentation products of Kribbella karoo-nensis A01098 were separated and purified by silica gel chromatography and Sephadex LH-20 gel column chromatography.The charac-terization of the isolated compounds was performed using nuclear magnetic resonance(NMR)and mass spectrometry(MS)to identify their structures.The inhibitory effects of the compounds on the proliferation of human liver cancer cells(HepG2)and cervical cancer cells(HeLa)were evaluated using the methylthiazolyldiphenyl-tetrazolium bromide(MTT)assay.Results Totally five compounds were isolated from the ethyl acetate fraction of the liquid fermentation products of Kribbella karoonensis A01098,and identified as cyclo-(L-proline-glycine),cyclo-(trans-4-hydroxy-L-proline-L-leucine),cyclo-(cis-4-proline-D-proline-L-phenylalanine),cyclo-(trans-4-hydroxy-L-proline-L-phenylalanine),and 2-aminobenzoic acid.The half-maximal inhibitory concentrations(IC50)of five compounds against HepG2 and HeLa cells were all higher than 50 μmol/L.Conclusion Five compounds,four cyclic dipeptides and one anthra-nillic acid,are isolated and identified from Kribbella karoonensis for the first time.The five compounds have no significant inhibitory effects on the proliferation of HeLa and HepG2 cells.
