摘要

Abstract

Objective To investigate the alleviating effect and mechanism of notoginsenoside R1(NGR1)on temporomandibular joint pain(TMJP)in mice and analyze the effect of NGR1 on gut flora.Methods C57BL/6J mice were randomly divided into 5 groups(n=12):sham group,TMJP group,12.5NGR1 group,25NGR1 group and 50NGR1 group.The mice in sham group were injected with 10 μL of normal saline into the bilateral temporomandibular joint(TMJ)cavity.The mice in the other groups were injected with complete Freund's adjuvant(5 mg/mL)into the bilateral TMJ cavity to induce the TMJP mouse model.One hour after the CFA injection,the mice in 12.5NGR1 group,25NGR1 group and 50NGR1 group were intraperitoneally injected with NGR1 at a dose of 12.5,25 and 50 mg/kg,respectively.The mice in the other groups were intraperitoneally injected with normal saline.The mice in each group were in-jected once a day for 4 consecutive days.At 24 h after the end of the administration,the pain threshold of facial mechanical stimulation was measured.HE staining was used to evaluate the morphology of the temporomandibular joint cartilage tissue,and toluidine blue(TB)staining was used to detect the level of proteoglycan in the temporomandibular joint cartilage tissue.The expressions of matrix metalloproteinase 13(MMP13)and interleukin-1β(IL-1β)in temporomandibular joint cartilage were detected by immunohistochemical staining.Serum levels of IL-1β,tumor necrosis factor-α(TNF-α)and IL-6 were detected by enzyme-linked immunosorbent assay(ELISA).The expression of Iba-1 and glial fibrillary acidic protein(GFAP)in brain tissue was detected by immunofluorescence staining.The mRNA and protein expression levels of inducible nitric oxide synthase(iNOS),IL-1β,TNF-α,IL-10 and arginase-1(Arg-1)in brain tissue were analyzed by RT-qPCR and Western blot.The contents of Bacteroidetes and Lachnospiraceae in stool samples were analyzed by RT-qPCR.Results Compared with sham group,the facial mechanical stimulation pain threshold of mice was de-creased in TMJP group(P<0.05),the temporomandibular joint showed obvious lesions,the proteoglycan content was decreased(P<0.05),the levels of MMP13 and IL-1β were increased(P<0.05),the levels of TNF-α,IL-1β and IL-6 were increased(P<0.05),the relative fluorescence intensity of Iba-1 was increased(P<0.05),the levels of iNOS,IL-1β and TNF-α were increased(P<0.05),and the contents of Bacteroidetes and Lachnospiraceae were decreased(P<0.05).Compared with TMJP group,the facial mechanical stimu-lation pain threshold of mice was increased in 12.5NGR1,25NGR1 and 50NGR1 groups(P<0.05),TMJ lesions were reduced,proteo-glycan content was increased(P<0.05),the levels of MMP13 and IL-1β were decreased(P<0.05),the levels of TNF-α,IL-1β and IL-6 were decreased(P<0.05),the relative fluorescence intensity of Iba-1 was decreased(P<0.05),the levels of iNOS,IL-1β and TNF-α were decreased(P<0.05),and the contents of Bacteroidetes and Lachnospiraceae were increased(P<0.05).Compared with 12.5NGR1 group and 25NGR1 group,the above measured indicators in 50NGR1 group were significantly different(P<0.05).Conclusion NGR1 can effectively alleviate TMJP,inhibit the expression of M1 markers and promote the expression of M2 markers in microglia,and improve the structure of gut flora.
