类风湿关节炎与男性生殖器癌发病风险的关联性:孟德尔随机化分析OA
Mendelian randomization analysis of relationship between rheumatoid arthritis and male genital cancer
目的 基于孟德尔随机化的研究方法探究类风湿关节炎与男性生殖器癌的关联性.方法 根据全基因组关联研究(GWAS)中单核苷酸多样性(SNP)的相关数据,基于孟德尔随机化的基本假设,以逆方差加权法(IVW)为主要分析方法以探寻类风湿关节炎与男性生殖器癌的因果关系,使用简单模型法、加权中位数法、加权模型法、MR-Egger回归法为补充.使用Cochran's Q检验、MR-Egger回归截距法和留一法进行敏感性分析.结果 类风湿关节炎与男性生殖器癌发病有因果关系,IVW法显示,类风湿关节炎与男性生殖器癌发病风险呈正相关(ID:ebi-a-GCST90018910,OR=1.001,95%CI:1.001~1.002,P<0.05;ID:ebi-a-GCST90038685,OR=1.221,95%CI:1.030~1.446,P<0.05).敏感性分析未发现异质性和水平多效性,留一法检验提示结果稳定.结论 类风湿关节炎可以使男性生殖器癌发病风险增加.
Objective To investigate the relationship between rheumatoid arthritis(RA)and male genital cancer using Mendelian ran-domization.Methods Based on the data of single-nucleotide polymorphism(SNP)from genome-wide association studies(GWAS),the causal relationship between RA and male genital cancer was investigated by inverse variance weighted(IVW)method,supple-mented with simple model method,weighted median method,weighted model methods,and MR-Egger regression method.The accu-racy of the results was verified through sensitivity tests including Cochran's Q test,MR-Egger intercept assessment and leave-one-out analysis.Results There was a causal relationship between RA and the incidence of male genital cancer.IVW results revealed a posi-tive correlation between RA and the risk of male genital cancer(ID:ebi-a-GCST90018910,OR=1.001,95%CI:1.001-1.002,P<0.05;ID:ebi-a-GCST90038685,OR=1.221,95%CI:1.030-1.446,P<0.05).Sensitivity analysis did not reveal heterogeneity or hori-zontal multicollinearity,and the leave-one-out test indicated stable results.Conclusion RA may increase the risk of male genital cancer.
