摘要

Abstract

Objective To study the effect and mechanism of AstragalosideⅣ(AⅣ)on the inflammatory response in Endometriosis(EMs)model mice.Methods A total of 100 female ICR mice were randomly divided into EMs study group(80 mice)and control group of(20 mice).The EMs model was established in the study group,and the sham surgery was performed in the control group.At 5 weeks after surgery,the study group was divided into 4 groups by random number table method,which were fed with different doses of AⅣ 0,5,10,and 30 mg/(kg·d),respectively.At 13 weeks after surgery,the peritoneal surgical site tissues of the study group and the control group were collected.HE staining was used to evaluate the degree of modeling and inflammatory response,and Western blot was used to evaluate the expression of TLR4 and NF-κB factors.The expression of interleukin(IL)-1β,IL-6,serum C-C motif chemokine ligand 2(Ccl-2)and tumor necrosis factor-α(TNF-α)was assessed by qRT-PCR and ELISA,followed by intervention with TLR4 inhibitor TAK-242.The expression changes of TLR4,NF-κB,IL-1β,IL-6,Ccl-2 and TNF-α in the epithelial cells of EMs mice were evaluated.Results Compared with the control group,with the increase of AⅣ dose,the protein expression levels of TLR4 and NF-κB,and the mRNA expression levels of IL-1β,IL-6,Ccl-2 and TNF-α in the study group decreased(P<0.05,P<0.01,P<0.001).After treatment with different doses of AⅣ and TAK-242,the expressions of TLR4 and NF-κB in the epithelial cells of the study group were significantly decreased,and the mRNA expressions of IL-1β,IL-6,Ccl-2 and TNF-α were decreased in a dose-dependent manner(P<0.05,P<0.01,P<0.001).Conclusion AⅣ can significantly reduce the inflammatory response of EMs in mice,which may be achieved through the mechanism of TLR4/NF-κB signaling pathway.
