Abstract

Bacteria are key factors in causing infections,and quantitative detection for bacteria is crucial for clinical monitoring and treatment guidance.Gold nanorods(Au NRs)were self-assembled into dense metallic films using a liquid-liquid interface method and successfully transferred onto an ultrathin thermoplastic polyurethane surface,forming a uniform and stable flexible surface-enhanced Raman scattering(SERS)active substrate.This substrate was modified with 4-mercaptophenylboronic acid(MPBA)to provide a reference signal for bacterial fixation.On this substrate,the detection limit for MPBA molecules can reach 1×10-9 mol/L.A gold-silver core-shell structured SERS tag was prepared,using 5,5'-dithiobis(2-nitrobenzoic acid)(DTNB)in the interlayer as the target signal.The signal of SERS tags can be enhanced by about 54 times by the prepared substrate.By combining SERS tags with nucleic acid aptamers,specific recognition of Escherichia coli(E.coli)was achieved.By analyzing the ratio of Raman characteristic peak intensities between the target signal and the reference signal(I1337/I1078),a good linear relationship was observed between bacterial concentration in the range of 1×102 to 1×107 CFU/mL and I1337/I1078 in water and saliva samples,with a coefficient of determination(R2=0.97126).It was superior to analyzing the target signal alone(R2=0.87046),demonstrating that ratiometric analysis provided a better fit.This strategy shows potential application value in bacterial detection.
