Preparation and Application of a Colloidal Gold Strip for Detecting Antibodies Against Schistosoma japonicum in Domestic Animals

Abstract
In order to rapidly and accurately detect antibodies against Schistosoma japonicum in livestock,a kind of colloidal gold strip was prepared through spraying recombinant streptococcal G protein(r-SPG)labeled by colloidal gold onto a glass cellulose membrane as a gold-labeled pad,and encapsulating soluble egg antigens(SEA)and anti-His tag mouse monoclonal antibodies(His-tag mAb)onto nitrocellulose membrane as a detection line and a quality control line,respectively,then the sensitivity,specificity,reproducibility,compliance rate and clinical application were evaluated.The results showed that the strip was with 96.43%sensitivity of diagnosis,failed to crossly react with the positive sera of Ornithobilharzia turkistanicum,Haemonchus contortus and Fasciola gigantica,and with good intra-and inter-batch repeatability.It could complete detection in 5-10 minutes.1 500 bovine and ovine sera were clinically tested,the total compliance rate of the strip with Schistosoma japonicum miracidium hatching method was found to be up to 98.1%.In conclusion,the strip was with high sensitivity,good specificity and reproducibility,and could strongly support the prevention and control of livestock Schistosomiasis japonicum in China.关键词
