血府逐瘀汤对黑素瘤小鼠放疗增敏作用及HDAC7/c-Myc通路调节作用研究
Effect of Xuefu Zhuyu Decoction on Radiotherapy Sensitization and Regulation of HDAC7/c-Myc Pathway in Melanoma Mice

Abstract
Objective To study the radiosensitization effect of Xuefu Zhuyu Decoction on melanoma mice and the regulation of HDAC7/c-Myc pathway.Methods The melanoma mouse model was established by armpit injection of B16-F10 cells,and divided into melanoma model group,Xuefu Zhuyu decoction low-dose,medium-dose and high-dose groups,radiotherapy group and Xuefu Zhuyu decoction combined radiotherapy group,10 mice per group.The low,medium and high dose groups of Xuefu Chuyu Decoction were respectively given the dose of 12.5 g/kg,25 g/kg and 50 g/kg by intragastric administration,once a day;the radiotherapy group was received radiotherapy at the dose of 3Gy for 2 times,and then was given radiotherapy at 4Gy.Xuefu Zhuyu Decoction combined radiotherapy group was treated with both radiotherapy and Xuefu Zhuyu Decoction,intervention for 6w.The serum levels of VEGF,IL-17 and IL-6 of the mice were measured by enzyme-linked immunosorbent kit,The tumor volume,tumor inhibition rate,tumor microvascular density,tumor cell apoptosis rate,HDAC7,c-myc mRNA and protein levels were measured.Results Compared with melanoma model group,serum VEGF,IL-17,IL-6 levels,tumor volume,tumor microvascular number,HDAC7,c-myc mRNA and protein levels of mice in Xuefu Zhuyu Decoction dose group,radiotherapy group and Xuefu Zhuyu Decoction combined radiotherapy group were significantly decreased(P<0.05).Tumor inhibition rate and tumor cell apoptosis rate were significantly increased(P<0.05),and with the increase of the dose of Xuefu Zhuyu Decoction,the changes of all indexes were more significant.Compared with Xuefu Zhuyu decoction high-dose group and radiotherapy group,serum VEGF,IL-17,IL-6 levels,tumor volume,tumor microvascular number,HDAC7,c-myc mRNA and protein levels of tumor tissue in Xuefu Zhuyu Decoction combined radiotherapy group were significantly decreased(P<0.05).Tumor inhibition rate and tumor cell apoptosis rate were significantly increased(P<0.05).Conclusion Xuefuzhuyu Decoction can significantly inhibit the tumor growth of melanoma mice,promote the apoptosis of melanoma tissue cells in mice,inhibit the formation of tumor microvessels,regulate the inflammatory response of melanoma mice,and then inhibit the progression of melanoma disease in mice.Meanwhile,Xuefuzhuyu Decoction can significantly enhance the radiotherapy effect of melanoma mice and improve the radiotherapy sensitivity of melanoma mice,and the mechanism may be related to the regulation of the HDAC7/c-Myc pathway.关键词
