Scanning Electron Microscope Observation and Phylogenetic Analysis of Three Species of Nematodes in the Strongylinae

Abstract
To analyze the morphological characteristics and phylogenetic relationships of three species of nematodes in the Strongylinae,this study used scanning electron microscope(SEM)to observe and measure the surface structures of Triodontophorus brevicauda(Boulenger,1916),Triodontophorus tenuicollis(Boulenger,1916),and Bidentostomum ivaschkini(Tshoijo,1957),which parasitize the intestines of equid animals.The ribosomal internal transcribed spacer(ITS)gene of these nematodes was PCR-amplified and sequenced,followed by homologous analysis and phylogenetic tree construction.SEM observations revealed that T.brevicauda had a short external leaf crown with small spines,with 46 spines in males and 64 in females.The male tail had a serrated copulatory bursa edge with one pair of linear copulatory spines and a barb at the tip.T.tenuicollis had a short and spiny external leaf crown,with 45 spines in males and 49 in females.The male tail also had a serrated copulatory bursa edge with one pair of linear copulatory spines and a barb at the tip,and the reproductive cone appendage was underdeveloped.B.ivaschkini had a wide external leaf crown with 8 spines in both males and females,and a small internal leaf crown.The male copulatory bursa was long with a smooth edge,the dorsal leaf was narrow and elongated,and the reproductive cone was extremely long with three sac-like protrusions at the tip.Phylogenetic analysis showed that the nematodes in the genus Triodontophorus and Bidentostomum were closely related but independent of each other,while the genus Triodontophorus and Bidentostomum was more distantly related to the genus Strongylus.T.tenuicollis was closely related to T.nipponicus,and T.brevicauda was closely related to T.serratus.These results provide detailed morphological data for these three nematode species and describe their phylogenetic relationships using molecular analysis.关键词
