Abstract

Objective To explore the mechanism of Astragali Radix-Dioscoreae Rhizoma and Puerariae Lobatue Radix-Salviea Miltiorrhizae Radix et Rhizoma in the treatment of type 2 diabetes mellitus(T2DM)complicated with coronary artery disease(CAD)based on network pharmacology and molecular docking.Methods By using TCMSP,UniProt and integrating literature reports,the chemical components and corresponding targets of Astragali Radix-Dioscoreae Rhizoma and Puerariae Lobatue Radix-Salviea Miltiorrhizae Radix et Rhizoma were retrieved.A Chinese materia medica-active components-target network was constructed using Cytoscape 3.9.1 software.Disease targets were screened through GeneCards,DurgBank,DisGeNET,OMIM and TTD databases.Venny 2.1.0 online platform was used to obtain the intersection targets between active components and diseases.STRING database was utilized to construct a protein-protein interaction network for the intersecting targets and identify core targets.By using Metascape database,GO function and KEGG pathway enrichment analysis were performed on intersection targets,and a Chinese materia medica-disease-active components-target-pathway network was constructed,and core active components were screened.PubChem database,PDB database,PyMOL 2.6.0 and AutoDockTools 1.5.6 software were used for molecular docking verification.Results A total of 93 active components were obtained,the core active components included quercetin,luteolin and puerarin.The PPI network showed that IL6,AKT1 and IL1Bwere the core targets.The results of molecular docking showed that the core components had good binding activity to the core target.Conclusion Astragali Radix-Dioscoreae Rhizoma and Puerariae Lobatue Radix-Salviea Miltiorrhizae Radix et Rhizoma can treat T2DM with CAD through AGE-RAGE,lipid and atherosclerosis,NF-κB signaling pathways,which may be related to inhibiting the secretion of inflammatory factors and regulating the metabolism of glucose and lipid.
