基于数据挖掘和网络药理学探讨中医药治疗原发性肝癌用药规律和作用机制
Exploration on the Medication Law and Mechanism of TCM Treatment for Hepatocellular Carcinoma Based on Data Mining and Network Pharmacology

Abstract
Objective To explore the medication law of TCM for the treatment of hepatocellular carcinoma(HCC)and the mechanism of its core drugs.Methods Medical cases of TCM treatment for HCC from January 1,2010 to December 31,2022 were retrieved from CNKI,Wanfang Data and VIP.The frequency statistics and complex network analysis of TCM were conducted using Ancient and Modern Medical Case Cloud Platform.Potential targets of TCM in treating HCC were predicted by utilizing TCMSP,Chinese Materia Medica Group Identification Database and the DrugBank database.Disease targets of HCC were predicted using GeneCards.Protein-protein interaction networks were constructed and core targets were screened using the STRING database and Cytoscape3.7.2 software.GO function and KEGG pathway enrichment analyses were performed using R4.2.3.Core drug active component-target-pathway networks were built using Cytoscape3.7.2 software.Results A total of 123 prescriptions were involved,including 182 kinds of Chinese materia medica.High frequency herbs were Scutellariae Barbatae Herb(39 times),Bupleuri RadixBupleurum chinense(37 times)and Hedyotis diffusa(33 times).The properties were mainly cold and cool,the main tastes were bitter and sweet,and the main meridians were liver,spleen,lung,heart and kidney meridians.Complex network analysis revealed that the core drugs were Scutellariae Barbatae Herb,Bupleuri Radix,Crataegi Fructus,Hedyotis diffusa,Eupolyphaga Steleophaga and Persicae Semen.There were 108 intersection targets between core drugs and HCC,with core targets including TP53,AKT1,VEGFA,EGFR,MYC,CASP3 and TNF.The potential signaling pathways for core drugs in treating HCC were the IL-17 pathway.The main active components of core drugs were quercetin,kaempferol,luteolin,wogonin,isorhamnetin,and β-sitosterol.Conclusion The treatment of HCC with TCM focuses on regulating liver qi,clearing heat and detoxifying,tonifying the spleen and enhancing qi,and promoting blood circulation to remove stasis.The main active components of core drugs act on the IL-17 signaling pathway to inhibit the progression of HCC.关键词
