Abstract
Objective To investigate the effect of miR-30a-5p on the expression of Human Epidermal Growth Factor Receptor 2(HER2)gene and Tumor Metastasis Suppressor Gene 1(TMSG-1)protein in breast cancer cells.Methods Human breast cancer MCF-7 cells were divided into the Mm group(MCF-7 cells transfected with miR-30a-5p mimic),the Nc group(MCF-7 cells transfected with miR-30a-5p negative control),and the Mc group(MCF-7 cells).Reverse Transcription-Poly-merase Chain Reaction(RT-PCR)and Western blot were used to detect the changes in HER2 and TMSG-1 levels and protein expression.Cell migration ability was detected by the transwell assay,MCF-7 cell proliferation was detected by the tetrazolium salt colorimetric assay,and MCF-7 cell apoptosis was detected by flow cytometry.Results There was no significant differ-ence in HER2 and TMSG-1 levels between the Nc group and the Mc group(P>0.05).Compared with the Nc group and the Mc group,HER2 levels in the Mm group were decreased,and TMSG-1 levels were increased(P<0.05).There was no significant difference in HER2 and TMSG-1 protein expression between the Nc group and the Mc group(all P>0.05).Compared with the Nc group and the Mc group,HER2 expression in the Mm group was decreased,and TMSG-1 expression was increased(all P<0.05).There was no significant difference in the apoptosis rate of MCF-7 cells between the Nc group and the Mc group(all P>0.05).The apoptosis rates of MCF-7 cells in the Nc group and the Mc group were(7.43±0.92)%and(7.40±1.03)%,respectively,while the apoptosis rate in the Mm group was(23.63±3.15)%,which was significantly higher than that in the Nc group and the Mc group(all P<0.05).There was no significant difference in the migration ability of MCF-7 cells between the Nc group and the Mc group(P>0.05),but the migration ability of MCF-7 cells in the Mm group was significantly lower than that in the Nc group and the Mc group(P<0.05).There was no significant difference in the proliferation ability of MCF-7 cells between the Nc group and the Mc group(P>0.05).Compared with the Nc group and the Mc group,the cell viability of the Mm group was weakened,and the prolif-eration ability of MCF-7 cells was significantly different among the three groups(all P<0.05).Conclusion Overexpression of miR-30a-5p leads to substantial apoptosis in MCF-7 cells,and its mechanism of action may be achieved by reducing HER2 and increasing TMSG-1 levels.关键词
