Relationship between Gingival Crevicular Fluid Cx43,TIMP-1 Levels and the Outcome of Root Canal Treatment in Patients with Irreversible Pulpitis

Abstract
Objective To explore the relationship between gingival crevicular fluid connexin 43(Cx43)and tissue inhibitor of metalloproteinases-1(TIMP-1)levels and the outcome of root canal treatment in patients with irreversible pulpi-tis(IP).Methods A total of 153 IP patients who underwent root canal treatment between January 2021 and June 2022(IP group)and 80 healthy volunteers(control group)were selected.IP patients were further divided into the unfavorable outcome group(31 cases)and the favorable outcome group(122 cases)based on treatment results.Enzyme-linked immunosorbent assay(ELISA)was used to measure Cx43 and TIMP-1 levels in gingival crevicular fluid.Logistic regression analysis was performed to identify factors influencing treatment outcomes,and receiver operating characteristic(ROC)analysis was conducted to evaluate the predictive performance of Cx43 and TIMP-1 levels.Results Cx43 and TIMP-1 levels were significantly higher in the IP group compared to the control group(P<0.05).High levels of Cx43 and TIMP-1 were independent risk factors for poor progno-sis after root canal treatment(P<0.05).The area under the ROC curve for combined Cx43 and TIMP-1 levels in predicting poor prognosis was 0.880,higher than for Cx43 or TIMP-1 alone(0.789 and 0.796,respectively;P<0.05).Conclusion Elevated levels of Cx43 and TIMP-1 in gingival crevicular fluid are associated with poor outcomes after root canal treatment in IP pa-tients,with high predictive performance when combined.关键词
