基于肠道菌群探讨大黄治疗慢性肾病的机制
Mechanism Exploration of Rhubarb in the Treatment of Chronic Kidney Disease Based on Gut Microbiota

Abstract
Objective:To explore the effects of Rhubarb on gut microbiota in chronic kidney disease(CKD)and to investigate its mechanisms in the prevention and treatment of CKD.Methods:A CKD mouse model was established using a 0.2%adenine diet,and Rhubarb decoction(2 g/kg)was administered by gavage for 14 days.Renal function changes were assessed by measuring plasma creatinine and urea levels.Pathological damage to the kidneys and intestines was examined using HE staining,while Masson staining was used to observe renal fibrosis.qRT-PCR was employed to detect the transcription levels of renal inflammation,fibrosis factors,and intestinal barrier factors.The structure of the gut microbiota was analyzed using 16S rDNA sequencing.Results:After Rhubarb intervention,the rate of body weight loss in CKD mice slowed,renal index increased,plasma creatinine levels decreased,and colon length increased(P<0.05).Additionally,structural damage to the kidneys was reduced,fibrosis was alleviated,and intestinal barrier integrity was improved.High-throughput 16S rDNA sequencing showed that Rhubarb treatment increased the abundance of beneficial bacteria such as Lachnospiraceae UCG-006 and Lachnospiraceae NK4A136 group,while reducing the abundance of harmful bacteria such as Alistipes,Rikenellaceae RC9 gut group,and Odoribacter.Changes in the gut microbiota were significantly correlated with inflammation,fibrosis,and intestinal barrier factors.Conclusion:Rhubarb decoction improves renal inflammation and fibrosis and enhances intestinal barrier integrity by modulating gut microbiota,thereby delaying the progression of CKD.关键词
