万氏愈瘫汤对缺血性脑卒中大鼠的治疗作用及机制探讨
Therapeutic Effect and Mechanism of Wanshi Yutan Decoction on Ischemic Stroke in Rats

Abstract
Objective:To investigate the effects of Wanshi Yutan Decoction on ischemic stroke in rats and explore its potential mechanisms.Methods:Forty-two Sprague-Dawley(SD)rats were randomly divided into seven groups:normal,sham-operated,model,low-dose,medium-dose(25.20 g·kg-1·d-1)and high-dose(50.40 g·kg-1·d-1)Wanshi Yutan Decoction(12.60 g·kg-1·d-1)groups,and positive drug(nimodipine 9.45 mg·kg-1·d-1)group,with six rats in each group.An ischemic stroke rat model was established,and rats were administered treatment accordingly.General behaviors were observed,and neurological function was assessed using the Modified Neurological Severity Score(mNSS).Brain tissue samples were collected,and TTC staining was used to observe infarct regions and calculate the infarction rate.Apoptosis in hippocampal tissue was evaluated using TUNEL staining.Western blot analysis was used to measure the expression levels of Cleaved-caspase-3,Bcl-2,and Bax in infarcted brain tissue.Results:Rats in the model group exhibited symptoms such as coarse fur,salivation,and poor mental state,with significantly increased mNSS scores(P<0.05),increased apoptosis in hippocampal tissue(P<0.05),elevated levels of Cleaved-caspase-3 and Bax(P<0.05),and reduced Bcl-2 expression(P<0.05).Compared to the model group,rats in the Wanshi Yutan Decoction groups showed symptom improvement(P<0.05),decreased mNSS scores(P<0.05),reduced apoptosis in hippocampal tissue(P<0.05),significantly lower levels of Cleaved-caspase-3 and Bax(P<0.05),and higher Bcl-2 expression levels(P<0.05).Conclusion:Wanshi Yutan Decoction may alleviate symptoms of ischemic stroke in rats,potentially through a mechanism related to the reduction of cell apoptosis in brain tissue.关键词
