Study on the HPLC Fingerprint and Chemical Pattern Recognition of Wild Chrysanthemum

Abstract
Objective:To establish the High-Performance Liquid Chromatography(HPLC)fingerprint of wild chrysanthemum and evaluate the quality of wild chrysanthemum from different origins and batches using chemical pattern recognition.Methods:The study utilized a Waters Symmetry C18 column(4.6 mm×250 mm,5 μm)with a mobile phase of acetonitrile-0.1%phosphoric acid aqueous solution for gradient elution.The flow rate was set at 0.6 mL·min-1,the column temperature at 25℃,and the UV detection wavelength at 334 nm,with an injection volume of 10 μL.Various chemical pattern recognition methods were used for the overall quality evaluation of multiple batches of wild chrysanthemum.Results:The HPLC fingerprints of 15 batches of wild chrysanthemum identified 18 common peaks,with similarities all greater than 0.9.Eleven common peaks,including linarin and neochlorogenic acid,were identified.Using Hierarchical Cluster Analysis(HCA),Principal Component Analysis(PCA),and Orthogonal Partial Least Squares-Discriminant Analysis(OPLS-DA),the 15 batches of wild chrysanthemum were categorized into three major groups,identifying two principal components and three differential quality markers:isochlorogenic acid A,isochlorogenic acid C,and linarin,which can be used as quality control indicators for wild chrysanthemum.Conclusion:The successfully established HPLC fingerprint method for wild chrysanthemum is simple,accurate,and has good reproducibility and reliability.It provides a reliable reference for the quality control and clinical application of wild chrysanthemum medicinal materials and preparations.关键词
