Abstract
To investigate the role and mechanism of α-1,3/1,6-mannosyltransferase Alg2 in enhancing the radiosensitivity of lung cancer cells,human non-small cell lung cancer cells(A549)were used to construct Alg2 overexpression and knockdown cell lines.Under irradiation conditions,the DR-GFP plasmid reporter system was used to detect the effect of Alg2 on DNA damage repair methods such as homologous recombination(HR)repair and non-homologous end joining(NHEJ)repair.The response of Alg2 knockdown cells to radiation was investigated by Western blot,immunofluorescence and other methods,and the regulatory mechanism of Alg2 on Ku70 glycosylation was further explored.The results showed that irradiation caused a significant increase in tumor cell death in Alg2 knockdown tumor cells;Alg2 knockdown significantly inhibited the non-homologous end joining(NHEJ)repair pathway,and Alg2 knockdown led to a decrease in Ku70 protein glycosylation.In conclusion,Alg2 inhibited the glycosylation of Ku70,a key protein in NHEJ repair,and after Alg2 inhibition,Ku70 chromatin recruitment was inhibited,which affected NHEJ repair in tumor cells and ultimately increased tumor cell radiosensitivity.关键词
