摘要

Abstract

[Objective]To screen microorganisms which had high tolerance to heavy metals,promoted plant growth and effectively blocked the transfer of heavy metals to the aboveground part of leafy vegetables,which could provide high-quality microbial resources for bioremediation of heavy metal contaminated soil.[Method]Bacillus was used as the test strain,which was screened for lead(Pb)resistant strains through selective media,and the maximum tolerance con-centration of Pb was determined using the dilution plating method.Different concentrations of Pb2+(25,50,100,200,400 mg/L)solutions were set up to analyze the strain's adsorption capacity for Pb using atomic absorption spectrometry.Germination experiments were conducted to analyze the growth promoting effects of the strain on the germination of Bras-sica chinensis seeds.Hydroponic experiments with different concentrations of Pb2+(0.3,30 mg/L)were conducted to in-vestigate the ability of the strain to block the absorption of Pb in the aboveground parts of Brassica chinensis.[Result]One strain of Bacillus subtilis was screened on LB solid medium with 1800 mg/L Pb2+concentration,which could grow normally and produce siderophore.Under conditions of natural pH,Pb2+initial concentration of 50 mg/L,wet strain con-centration of 16 g/L,temperature of 30℃,rotation speed of 150 r/min and adsorption time of 24 h,the strain achieved the highest Pb adsorption rate up to 96.1%.The seed germination experiment results showed that the Bacillus subtilis in-creased the germ length and root length of Brassica chinensis seeds under both Pb stress and non-stress conditions.The re-sults of the hydroponic experiment showed that,compared to non-inoculated treatments,at a higher Pb2+concentration(30 mg/L),the fresh weight and dry weight of B.chinensis treated with Bacillus subtilis increased significantly by 148.8%and 125.0%respectively(P<0.01),the inoculation treatments with bacterial strains at Pb2+concentrations of 0.3 and 30 mg/L significantly reduced the Pb content in the aboveground parts of Brassica chinensis by 53.5%and 38.9%(P<0.05,the same below)respectively,the Pb transfer coefficient was also significantly decreased by 23.1%and 20.5%respectively.[Conclusion]A strain of Bacillus subtilis has been screened,which not only exhibits high tolerance and adsorption capacity for Pb,but also shows significant plant growth promoting functions and the ability to block Pb trans-fer to the aboveground parts of Brassica chinensis.
