沙枣果总黄酮对大鼠PCOS-IR模型的干预作用研究
Study on the intervention effect of total flavone of oleaster fruit on PCOS-IR model in rats

Abstract
Objective:To investigate the therapeutic effect of total flavone of oleaster fruit on the rat model of PCOS complicated with insulin resistance(PCOS-IR). Methods:The PCOS-IR model was established by HCG+insulin+high-fat and high-sugar diet.A blank group was established simultaneously.The rats with successful modeling were randomly divided into five groups:model group,metformin group(104 mg/kg),low dose flavone group(50 mg/kg),high dose flavone group(200 mg/kg)and metformin combined with high dose flavone group(104 mg/kg+200 mg/kg),with five rats in each group.Rats in the blank and the model group received equal volume normal saline.After continuous administration for 26 days,the samples were weighed and the relevant indexes were detected. Results:Compared with the model group,the pathological damages of ovarian tissue structure disorder,corpus luteum reduction,and the increase of the number of cystic follicles and atretic follicles improved with different degrees.At the same time,body mass,uterine index(except for low dose flavone group),ovarian index,fasting insulin level,the homeostatic model assessment for insulin resistance(HOMA-IR),testosterone(T)and LH decreased to varying degrees(P<0.05),while estradiol(E2,except for low dose flavone group),progesterone(P)and FSH levels increased(P<0.05),especially in metformin combined with high dose flavone group(P<0.01).Although the uterine index showed a certain downward trend and the E2 level showed an upward trend in low dose flavone group,the differences were not statistically significant when compared with the model group(P>0.05).Compared with high dose flavone group,there was a significant decrease in body mass,HOMA-IR and LH level(P<0.05),and a significant increase in the levels of E2,P and FSH in metformin combined with high dose flavone group(P<0.05).The levels of T,uterine index and ovarian index also changed,but the differences were not statistically significant(P>0.05). Conclusions:Total flavone of oleaster fruit had a certain therapeutic effect on PCOS-IR rat model,and the results of this study could provide preliminary experimental support for the treatment of PCOS-IR by total flavone of oleaster fruit.关键词
