Abstract
Objective:To investigate the effect of baicalein(Bai)on pregnancy out-comes and phosphatidylinositol 3-kinase/protein kinase B(PI3K/Akt)signaling pathway in rats with threatened miscarriage.Methods:Female pregnant mice were randomly grouped into Control group,Model group,Bai low-dose(Bai-L)group,Bai high-dose(Bai-H)group,posi-tive control progesterone group(PROG group),and Bai high-dose+PI3K inhibitor LY294002(Bai-H+LY294002)group.Except for the Control group,the threatened miscarriage model was constructed.The miscarriage rate of rats in each group was calculated.HE staining was used to detect the pathological changes of uterine tissue structure in each group.Enzyme-linked immu-nosorbent assay(ELISA)was used to detect the expression levels of serum interferon-γ(IFN-γ),tumor necrosis factor-α(TNF-α),interleukin-10(IL-10),β-human chorionic gonadotropin(β-HCG),progesterone,estradiol and other hormones in each group.Western blot was used to detect the expression level of PI3K/Akt signaling pathway-related proteins in uterine tissue.Re-sults:Compared with the Control group,the uterine tissue structure of rats in the Model group was damaged,with loose cell arrangement,dense stroma,and inflammatory infiltration in the en-dometrium,the miscarriage rate and the expression levels of IFN-γ,TNF-α and IL-10 in serum were greatly increased(P<0.05),the levels of estradiol,β-HCG,progesterone,and the p-PI3K/PI3K and p-Akt/Akt in uterine tissue were greatly decreased(P<0.05).Compared with the Model group,the degree of uterine tissue structure damage and endometrial inflammatory in-filtration in the Bai-L group and Bai-H group were reduced,and the cell arrangement gradually tended to be orderly,the miscarriage rate and the expression levels of IFN-γ,TNF-α and IL-10 in serum were gradually decreased(P<0.05),the levels of estradiol,β-HCG,progesterone,and the p-PI3K/PI3K and p-Akt/Akt in uterine tissue were gradually increased(P<0.05).There was no great difference between the Bai-H group and the PROG group(P>0.05).The addition of PI3K inhibitor LY294002 reversed the effects of Bai on the above indexes(P<0.05).Con-clusion:Bai can improve pregnancy outcomes in threatened miscarriage rats,and its mechanism of action may be related to the activation of the PI3K/Akt signaling pathway.关键词
