Abstract

To explore the function of African swine fever virus(ASFV)R298L gene,elucidate the pathogenic mechanism of ASFV,and provide a candidate strain for the vaccine research,the gene deletion ASFV-△R298L was constructed by homologous recombination.The pathogenicity and immune response of ASFV-△R298L and ASFV CN/GS/2018 were evaluated in pigs.The results showed that animals infected with ASFV-WT had severe fever and all died within 13 days after infection,which severe lesions occurred in multiple organs,but the animals infected with ASFV-△R298L had no fever or lesions in organs and all survived during the observation period.More importantly,pigs infected with ASFV-△R298L produced high levels of neutralizing antibody in late stage.In summary,these findings provide insights into analysis the pathogenic mechanism of ASFV,and provide important ideas for the development of gene deletion candidate vaccines.
